SOD (EC 1.15.1.1) is a kind of metal enzyme which widely exists in organisms. It is an important free radical scavenger and can catalyze the disproportionation of superoxide anion to produce H2O2 and O2. SOD is not only the superoxide an scavenging enzyme, but also the main generating enzyme of H2O2, which plays an important role in the biological antioxidant system.

The production of superoxideion (O2-) was through the system of xanthine and xanthine oxidase reaction, O2- can reduce the nitro blue tetrazolium to form blue formazan, which has an absorption at 560 nm; SOD can clear O2-, thus inhibited the formation of formazan; the deeper the blue the reaction solution, the lower the SOD activity, and vice versa the higher the activity.
