Significance of Determination

Proline (Pro) is widely present in animals, plants, microorganisms, and cultured cells. Under stress conditions, the content of Pro in plants increases significantly. The increase in Pro reflects stress resistance to a certain extent—varieties with strong drought resistance tend to accumulate more proline. Therefore, the increase in proline can be used as one of the physiological indicators for stress-resistant breeding.

Principle of Determination

Pro is extracted with sulfosalicylic acid (SA). After heat treatment, Pro reacts with acidic ninhydrin solution to form a red product, and the absorbance is measured at 520 nm.

Instruments and Supplies to Be Prepared by Yourself

Instruments: Visible spectrophotometer/microplate reader, water bath, desktop centrifuge, adjustable pipette, micro quartz cuvettes/96-well plates, mortar.

Supplies: 25 mL glacial acetic acid, ice, distilled water.
