Oxygen free radicals act on unsaturated fatty acids in lipids to generate lipid peroxides. The latter gradually decompose into a series of complex compounds, including malondialdehyde (MDA). The level of lipid oxidation can be determined by detecting the level of MDA.

Under acidic and high-temperature conditions, malondialdehyde (MDA) can condense with thiobarbituric acid (TBA) to form a reddish-brown trimethine (3,5,5-trimethyl oxazole-2,4-dione), whose maximum absorption wavelength is 532 nm. The content of MDA in the sample can be estimated after colorimetry.
