pKNT25 Vector

This vector is a derivative of the bacterial two-hybrid system (e.g., BACTH system), sharing core functions with pKT25 but differing in fusion mode. Its multiple cloning site is located downstream of the T25 gene, allowing insertion of target genes to construct an expression cassette where the target protein's N-terminus is fused with T25's C-terminus. After co-transformation with vectors carrying the T18 fragment, the interaction of fusion proteins enables T25 and T18 to reconstitute active adenylate cyclase, activating cAMP-dependent reporter gene expression to detect protein-protein interactions. It is suitable for target genes where N-terminal fusion may affect protein structure or function, providing an alternative fusion orientation to optimize the accuracy of prokaryotic protein interaction studies.
