The core principle of the Pichia pastoris transformation enhancer is to improve the efficiency of exogenous DNA entry through 'barrier disruption and assisted binding.' Commercial transformation enhancers often contain permeabilizing components that, when activated by boiling water bath, increase cell membrane permeability; lithium chloride disrupts the cell wall structure, reducing resistance to DNA entry; PEG3350 can neutralize the toxicity of high-concentration LiCl while promoting the binding of DNA to the cell surface; single-stranded salmon sperm DNA can adsorb exogenous DNA, assisting it to more easily enter yeast cells.
