Protein carbonylation refers to the process where amino or imino groups in the side chains of amino acid residues are attacked by oxygen free radicals, eventually converted into aldehyde groups, and release NH₃⁺. Its formation in vivo is mainly accomplished through the metal ion-catalyzed oxidation system.Protein carbonyls are early markers of oxidative modification of proteins involving multiple amino acids. The level of protein carbonyls indicates the degree of protein oxidative damage, and it is a key indicator for measuring protein oxidative damage.

Carbonyl groups react with 2,4-dinitrophenylhydrazine (DNPH) to form 2,4-dinitrophenylhydrazone, which has a characteristic absorption peak at 370 nm.
