pUT18C Vector
This vector is one of the core vectors of the bacterial two-hybrid system (e.g., BACTH system), carrying the coding sequence of the C-terminal domain (T18 fragment) of adenylate cyclase. The multiple cloning site is located upstream of the T18 gene, allowing insertion of target gene sequences to construct a fusion protein expression cassette with T18. When co-transformed into E. coli with vectors carrying the T25 fragment (N-terminal domain of adenylate cyclase) (such as pKT25), if the two fusion proteins interact, T18 and T25 can reconstitute active adenylate cyclase, catalyzing cAMP synthesis and activating downstream reporter genes (e.g., lacZ). Protein-protein interactions can be detected through phenotypic changes. It is suitable for prokaryotic protein interaction screening, interaction strength analysis, and functional verification.
