pCCW-Alg5 Vector
This vector is a specialized tool for the yeast split-ubiquitin two-hybrid system, primarily functioning as a positive control for detecting membrane protein interactions. It carries a fusion gene of the Alg5 protein C-terminal coding sequence and the ubiquitin C-terminal domain (Cub), driven by the yeast CCW12 promoter . As an endoplasmic reticulum transmembrane protein, Alg5 has its C-terminus localized in the cytoplasm, which can specifically interact with target proteins containing the ubiquitin N-terminal domain (NubI). This interaction reconstitutes a complete ubiquitin molecule and activates downstream reporter genes (e.g., HIS3, ADE2, LacZ), enabling yeast growth on nutrient-deficient media and corresponding color reactions . This vector is widely used to validate the efficacy of membrane protein interaction systems, analyze bait protein localization, and calibrate interaction signal thresholds. It is particularly suitable for studying interactions of hydrophobic proteins such as mitochondrial membrane proteins and G-protein-coupled receptors .
