pDL2-Alg5 Vector
his vector is a dedicated one for the yeast split ubiquitination-based two-hybrid system, primarily designed as a control tool for detecting membrane protein. It harbors the sequence of the Alg5 protein fused to the N-terminal domain (NubG) of a ubiquitin mutant, driven by the yeast CC12 promoter. NubG is unable to reconstitute the complete ubiquitin molecule with Cub and activate downstream reporter genes (such as HIS3, ADE2, Lac), thus failing to support yeast growth on a selective medium and exhibit the corresponding colorimetric reaction. This vector is widely used for validating the effectiveness of membrane protein interaction systems bait protein localization analysis, and calibration of interaction signal thresholds.
