Chalcone isomerase (CHI) is the first recognized enzyme involved in flavonoid biosynthesis, and it is also one of the key enzymes in the flavonoid metabolic pathway. Together with chalcone synthase (CHS), chalcone isomerase constitutes the rate-limiting enzymes in the biosynthesis of flavonoids.

Chalcone isomerase (CHI) catalyzes the cyclization of chalcone to form 4,5,7-trihydroxyflavanone. The activity of CHI is represented by measuring the change in absorbance at 381 nm.
