Polyamine Oxidases (PAO) are key enzymes that catalyze the oxidation of polyamines in organisms. Polyamines can interact with nucleic acids, proteins, and cell membrane molecules, directly affecting cell growth, differentiation, and apoptosis. By regulating the intracellular polyamine levels and the concentration of their metabolites, PAO participates in various plant responses to abiotic stresses and plant growth and development processes.

PAO catalyzes the oxidation of polyamines to generate hydrogen peroxide (H₂O₂). In the presence of oxygen, peroxidase catalyzes the decomposition of H₂O₂, and simultaneously oxidizes o-dianisidine to form a colored substance, which has a characteristic absorption peak at 500 nm. The intensity of the color shows a linear relationship with PAO activity.
