YM Medium
YM Medium is prepared according to the Wickerham formula, used for isolating and cultivating yeasts, molds, and other acidophilic bacteria. The addition of antifungal agents such as sodium propionate and biphenyl is used to inhibit mold growth, allowing yeast to be counted from mixed cultures. Wickerham recommended using yeast malt broth as an enrichment broth for yeasts by adding a layer of sterile paraffin oil (about 1 cm thick) on the surface of the medium. If there is growth in the medium, yeast colonies can be isolated on YM agar by streaking. To obtain fermentative and oxidative bacteria, acid can be added to the YM broth, followed by shaking incubation for 1–2 days. These conditions favor yeast growth while preventing molds from sporulating. Subsequently, yeasts can be isolated from YM agar plates.
