Alcohol dehydrogenase catalyzes the dehydrogenation of ethanol to acetaldehyde, while reducing NAD⁺ to generate NADH and H⁺. Under the action of 1-methoxyphenazine methosulfate (1-mPMS), WST-1 (2-(4-iodophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, monosodium salt) can react with NADH to produce water-soluble formazan, which has a maximum absorption peak at 450 nm. The ethanol content can be calculated based on this.
