CoenzymeⅡ NADP(H)Content Assay Kit
CoenzymeⅡ NADP(H)Content Assay Kit
Coenzyme Ⅱ, NADP(H), is widely present in animals, plants, microorganisms, and cultured cells. The determination of NADP⁺ and NADPH contents allows the calculation of total NADP content (NADPH + NADP⁺) and the NADPH/NADP⁺ ratio. Changes in these parameters are closely related to the pentose phosphate pathway (PPP), biosynthesis, and antioxidant reactions. The NADPH/NADP⁺ ratio is not only one of the main indicators of the cellular redox state but also plays an important regulatory role in the PPP pathway, biosynthesis, and antioxidant metabolism. NADP⁺ and NADPH in samples are extracted using acidic and alkaline extracts, respectively. Through the hydrogen transfer of PMS (phenazine methosulfate), NADPH reduces oxidized methyl thiazolyl tetrazolium (MTT) to formazan. The absorbance is measured at 570 nm to determine the NADPH content. Glucose-6-phosphate dehydrogenase is used to reduce NADP⁺ to NADPH, thereby determining the NADP⁺ content.
