Nitric oxide (NO) is widely distributed in systems such as the nervous, circulatory, respiratory, digestive, and genitourinary systems in organisms, and is particularly abundant in neural tissues. As an intercellular and intracellular signaling molecule, it plays a role in signal transmission and is a novel biological messenger molecule, exerting important effects in the physiological and pathological processes of the organism.

NO is highly prone to oxidation into nitrite ions (NO₂⁻) in vivo or in aqueous solutions. Under acidic conditions, NO₂⁻ reacts with sulfanilamide (a diazonium salt precursor) to form a diazonium compound, which further couples with N-(1-naphthyl) ethylenediamine. The resulting product has a characteristic absorption peak at 550 nm. By measuring its absorbance, the content of NO can be calculated.
