Glutamate-pyruvate transaminase (GPT, EC 2.6.1.2) is widely present in animals, plants, microorganisms, and cultured cells. It catalyzes the transamination reaction between amino acids and keto acids, playing an important role in amino acid metabolism. Additionally, the activity of GPT is very high in mammalian hepatocytes; when hepatocytes undergo necrosis, GPT is released into the blood, leading to a significant increase in serum GPT activity. Therefore, GPT has been recommended by the World Health Organization (WHO) as the most sensitive indicator for detecting liver function impairment.

GPT catalyzes the transamination reaction between alanine and α-ketoglutarate, producing pyruvate and glutamic acid. A 2,4-dinitrophenylhydrazine solution is added, which not only terminates the above reaction but also undergoes an addition reaction with the carbonyl group in keto acids to form pyruvate phenylhydrazone. The phenylhydrazone exhibits a reddish-brown color under alkaline conditions, and the absorbance can be measured at 505 nm to calculate the enzyme activity.
