Proline dehydrogenase (ProDH) is a key enzyme located in mitochondria that catalyzes the degradation of proline. Proline is one of the most widely distributed osmoprotectants; under stress conditions, many plants can accumulate large amounts of proline in vivo by increasing its synthesis and reducing its degradation. Reducing ProDH activity is of great significance for regulating osmotic balance, preventing osmotic stress from damaging plants, scavenging free radicals, and protecting cell structures.

Methyl isothiocyanate is used to detect the dehydrogenation reaction catalyzed by ProDH, and the change in absorbance at 600 nm reflects the level of ProDH activity.
