Lysine is one of the essential amino acids for humans. It can promote human growth and development, enhance immune function, and play a role in improving the function of central nervous tissue. Lysine is a basic essential amino acid. Due to its extremely low content in cereal foods and easy destruction (thus deficiency) during processing, it is known as the first limiting amino acid.

Lysine in proteins has a free ε-amino group (ε-NH₂), which reacts with ninhydrin reagent to form a blue-purple substance. Within a certain range, the depth of the color has a linear relationship with the content of lysine. Leucine has the same number of carbon atoms as lysine and only has one free amino group (without the ε-NH₂ group), so leucine is usually used to prepare the standard solution.
