Proline is an important osmotic adjustment substance for plants to adapt to stress conditions. In higher plants, proline metabolism is divided into two synthetic pathways—using glutamate (Glu) and ornithine (Orn) as initial substrates, respectively. δ-Ornithine aminotransferase (δ-OAT) is a key enzyme in the proline synthesis pathway that uses ornithine as a precursor, and plays a crucial role in helping plants adapt to stress conditions.

Under the action of δ-OAT and NADH, ornithine and α-ketoglutarate undergo a transamination reaction to generate pyrroline-5-carboxylate (P5C), with the simultaneous production of NAD⁺. The change in absorbance at 340 nm can be measured to reflect the level of δ-OAT activity.
