Proline is an important osmotic adjustment substance for plants to adapt to stress conditions. In higher plants, proline metabolism is divided into two synthetic pathways—using glutamate (Glu) and ornithine (Orn) as initial substrates, respectively. Δ1-Pyrroline-5-carboxylate synthetase (P5CS) is a key enzyme in the proline synthesis pathway that takes glutamate as the precursor, and plays a crucial role in helping plants adapt to stress conditions.

During the process where P5CS catalyzes the conversion of glutamate to pyrroline-5-carboxylate (P5C), it decomposes ATP to produce ADP and inorganic phosphate. The activity of P5CS can be determined by measuring the amount of inorganic phosphate generated per unit time using the ammonium molybdate colorimetric method.
