Cysteinyl Sulfoxide Lyase (CSL) Assay Kit

Cysteine sulfoxide lyase, commonly known as alliinase, is also called alliin lyase. It exists in almost all plants of the Allium genus, such as garlic, onions, and leeks. Alliinase is localized in vacuoles, while its natural substrate, alliin, is present in the cytoplasm. When alliinase comes into contact with alliin, it catalyzes the production of allicin and cis/trans-ajoene, along with by-products such as pyruvate and ammonia; this reaction is also the main source of the pungent odor in plants like garlic.

Cysteine sulfoxide lyase can catalyze S-methyl-L-cysteine sulfoxide to generate pyruvate. Pyruvate can react with 2,4-dinitrophenylhydrazine to form 2,4-dinitrophenylhydrazone, which exhibits a brownish-red color under alkaline conditions. The activity of cysteine sulfoxide lyase (CSL) can be calculated by measuring the change in absorbance at 505 nm.
