Glutamate Decarboxylase (GAD) Assay Kit
Glutamate Decarboxylase (GAD) Assay Kit
Glutamate decarboxylase (GAD, EC 4.1.1.15) is a highly specific pyridoxal phosphate-dependent amino acid decarboxylase, present in animals, plants, and microorganisms. GAD is the key enzyme that catalyzes the synthesis of γ-aminobutyric acid (GABA) in organisms. In the animal central nervous system, GABA functions as an inhibitory neurotransmitter and also plays an auxiliary therapeutic role in lowering blood pressure, improving brain function, and enhancing liver and kidney functions. In plants, GABA mainly exerts effects in resisting stress conditions such as low temperature, drought, and pest infestation, inducing ethylene synthesis, and regulating growth and development. Under the action of the coenzyme pyridoxal phosphate, GAD catalyzes the substrate glutamate to produce GABA and carbon dioxide. GABA then reacts with hypochlorite and phenols to form a blue-green substance, which has a characteristic absorption peak at 640 nm. By measuring the absorbance at 640 nm, the amount of GABA produced can be calculated, and the GAD activity in the sample can be further derived.
