Determination principle: Glucose oxidase (GOD) catalyzes the oxidation of glucose to gluconic acid, producing hydrogen peroxide (H₂O₂); peroxidase (POD) then catalyzes the oxidation of 4-aminoantipyrine-coupled phenol by hydrogen peroxide, forming a colored compound which has a characteristic absorption peak at 505 nm.
