GM2163
The GM2163 strain exhibits resistance to chloramphenicol and streptomycin sulfate, and is a K12 strain lacking methyltransferases dam and dcm. The extracted plasmid DNA can be cleaved by endonucleases sensitive to dam and dcm methylation. Mutations in methyltransferases dam and dcm lead to increased intracellular gene mutation rates, resulting in the formation of two colony sizes during growth. When isolating colonies, prioritize medium or smaller-sized colonies. GM2163 lacks laclqlacZ△M15, making blue-white spot screening infeasible. It does not carry endonuclease endA1 mutations but contains high levels of endonucleases in vivo. When extracting plasmids, it is essential to use the protein-dealing solution from the plasmid extraction kit to minimize endonuclease contamination of the plasmids.
