Carbohydrates are one of the important components of plants, and also serve as the main raw materials for metabolism and storage substances. Total sugars, also referred to as carbohydrates, include soluble monosaccharides, disaccharides, and insoluble substances such as starch, cellulose, and chitin.

Total sugars are hydrolyzed into reducing sugars by acid. In the presence of NaOH and glycerol, the DNS reagent reacts with reducing sugars under heating and is reduced to amino compounds. These compounds appear orange-red in an alkaline solution with excess NaOH and have a maximum absorption peak at 540 nm. Total sugar content in the sample is determined based on this principle.
