YPDS Liquid Medium（containing glucose and sorbitol）

YPDS medium, which is YPD with added sorbitol, is used in genetic transformation experiments of yeast (such as introducing foreign DNA yeast cells by electroporation or chemical methods). During these experiments, the yeast cell wall is damaged and becomes very fragile. If the transformed yeast is directly spread on a commonPD plate, due to the imbalance of osmotic pressure between the medium and the inside of the cell, the fragile cells will burst and die under the osmotic pressure shock Sorbitol, as an inert and metabolically difficult sugar alcohol, can increase the osmotic pressure of the medium, making it equal or close to the osmotic pressure the yeast cell. This provides a hypertonic and stable environment for the damaged yeast cells, protecting them from lysis and giving them the opportunity to repair the cell wall and start.
