Hemicellulose is a polysaccharide mixture tightly bound to cellulose in plant cell walls. It is a major component of the primary cell wall and widely present in plants, serving as a new type of usable energy source.

After acid treatment, hemicellulose is converted into reducing sugars. These reducing sugars react with 3,5-dinitrosalicylic acid (DNS) to form a reddish-brown substance, which has a characteristic absorption peak at 540 nm. The absorbance value reflects the content of hemicellulose.
