Fructose-1,6-Diphosphate (FDP) is an important intermediate in the glycolytic pathway. It regulates a variety of enzymes and has effects such as improving cellular energy metabolism, increasing energy utilization, anti-arrhythmia, and anti-tissue peroxidation. Thus, it is widely used in clinical medicine.

Aldolase catalyzes FDP to generate glyceraldehyde-3-phosphate and dihydroxyacetone phosphate. Then, under the action of triosephosphate isomerase and α-glycerophosphate dehydrogenase, nicotinamide adenine dinucleotide (reduced form, NADH) and dihydroxyacetone phosphate are catalyzed to form nicotinamide adenine dinucleotide (oxidized form, NAD⁺) and α-glycerophosphate, resulting in a decrease in absorbance at 340 nm. The content of FDP can be calculated by measuring the change in NADH.
