Glucose-6-phosphatase (G6Pase, EC 3.1.3.9) is a phosphatase that hydrolyzes phosphate compounds. It is widely present in animals, plants, microorganisms, and cells. As the rate-limiting enzyme in gluconeogenesis that hydrolyzes glucose-6-phosphate to produce glucose, it plays an important role in maintaining the dynamic balance of blood glucose.

G6Pase catalyzes glucose-6-phosphate to generate glucose and inorganic phosphate. The activity of G6Pase can be reflected by measuring the increase in inorganic phosphate content using the molybdenum blue method.
