pGreenII 0800-LUC-ccdb Vector 

pGreenII 0800-LUC-ccdb is based on the pGreenII 0800 backbone, with core elements including firefly luciferase (LUC) gene, ccdb negative selection gene and multiple cloning sites. The toxic protein encoded by ccdb gene can kill Escherichia coli without resistance gene. Only when the target fragment (such as promoter sequence) is inserted and replaces ccdb gene, the recombinant vector can survive in conventional Escherichia coli, realizing efficient positive clone screening. This vector clones the target promoter upstream of LUC, so that LUC expression is regulated by the target promoter. The transcriptional activity of the promoter is quantitatively reflected by detecting luciferase activity, which is suitable for analyzing plant promoter strength, studying the response characteristics of inducible promoters and verifying the function of transcriptional regulatory factors.
