Mitochondrial Malate Dehydrogenase (MDHm) Assay Kit
Mitochondrial Malate Dehydrogenase (MDHm) Assay Kit
Malate Dehydrogenase (MDH, EC 1.1.1.37) is widely present in animals, plants, microorganisms, and cultured cells. MDH in mitochondria is one of the key enzymes of the tricarboxylic acid (TCA) cycle, catalyzing the conversion of malate to oxaloacetate; conversely, MDH in the cytoplasm catalyzes the conversion of oxaloacetate to malate. Oxaloacetate is an important intermediate that connects multiple critical metabolic pathways. Therefore, MDH plays a vital role in various cellular physiological activities, including mitochondrial energy metabolism, the malate-aspartate shuttle system, reactive oxygen species metabolism, and disease resistance. Based on different coenzyme specificities, MDH is classified into NAD-dependent MDH and NADP-dependent MDH. Bacteria usually only contain NAD-MDH, while in eukaryotic cells, NAD-MDH is distributed in the cytoplasm and mitochondria. Mitochondrial MDH (MDHm) catalyzes the reduction of oxaloacetate by nicotinamide adenine dinucleotide (reduced form, NADH) to generate malate, which leads to a decrease in light absorption at 340 nm. The activity of MDHm can be determined by measuring the rate of this absorbance decrease.
