3-Hydroxy-3-methylglutaryl-CoA synthase (HMGCS) is a key enzyme in the mevalonate metabolic pathway. It catalyzes the reaction between acetyl-CoA and acetoacetyl-CoA to form 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA).

HMGCS catalyzes acetyl-CoA and acetoacetyl-CoA to generate HMG-CoA, with the simultaneous production of coenzyme A (CoASH). CoASH converts 5,5'-dithiobis-(2-nitrobenzoic acid) (DTNB) into yellow 2-nitro-5-thiobenzoic acid (TNB), which has a characteristic absorbance at 412 nm. The activity of HMGCS can be quantified by measuring the change in absorbance of TNB at 412 nm.
