Signal Peptide Secretion Yeast Detection Kit

The signal peptide secretion yeast detection kit is a tool used to verify the secretion function of signal peptides. It typically employs the pSUC2 yeast secretion system using the YTK12 yeast strain, which lacks the sucrose invertase gene and cannot grow on media where sucrose is the sole carbon source. The selected plasmid vector pSUC2 contains the tryptophan synthesis gene and a SUC2 invertase gene missing the signal peptide and start codon ATG. When the signal peptide to be tested is linked to the SUC2 invertase gene in pSUC2, if the signal peptide has secretion function, it can restore the secretion ability of invertase, allowing the YTK12 strain to grow on defective media with raffinose as the carbon source. This can be further verified using the TTC colorimetric reaction.
