pMAD Vector

pMAD is a gene knockout vector for Gram-positive bacteria such as Bacillus subtilis. It contains a temperature-sensitive replicon and carries a erythromycin resistance gene for selection. Its temperature-sensitive feature facilitates plasmid curing and it also includes a lacZα screening marker. It is commonly used to construct gene deletion mutants and is widely applied in the study of gene function in Bacillus species.
