pCVD442 Vector

pCVD442 is a commonly used suicide plasmid vector containing a ColE1 replicon (replicating only in Escherichia coli harboring the pir gene) and the sacB suicide gene (encoding levansucrase, which renders bacteria unable to grow on sucrose-containing medium). The vector is designed for gene editing in Gram-negative bacteria (such as Escherichia coli, Salmonella, etc.), enabling integration of target fragments into the host genome via homologous recombination, followed by elimination of the vector backbone using sucrose selection pressure. It is suitable for bacterial gene knockout, site-directed mutagenesis, and gene replacement studies, and serves as an important tool in microbial genetics and research on pathogenic mechanisms of pathogens.
