p3HR2 is a core vector of the yeast three-hybrid system, which is modified from the pIIIA/MS2-2 vector by deleting the ADE2 gene for simplification, and contains a unique RNA expression framework. Its key structures include a GC clamp (stabilizing RNA secondary structure), MS2 RNA stem-loop repeat sequences (mediating the binding of RNA to MS2 coat protein), and multiple restriction sites (XhoI, XmaI/SmaI, SphI), which facilitate the insertion of bait RNA sequences and ensure their correct display in the loop region to avoid the formation of inhibitory secondary structures . This vector is mainly used to screen for proteins that interact with target RNA. By cloning the bait RNA into the restriction sites and co-transforming it with the prey vector containing the candidate protein gene into L40coat yeast, positive interacting clones are screened using auxotrophic plates. It is widely used in studies such as the screening of viral subgenomic RNA-binding proteins and the functional verification of plant RNA-binding proteins .
