SS/-Ura with Agar

SS/-Ura medium is the same as SD medium except glucose is replaced by sucrose. The composition of SS/-Ura solid medium is the as SS/-Ura medium except that agar powder is added. The composition includes YNB (without amino acids and with ammonium sulfate), sucrose, agar, and 17 amino acids and nucleotide molecules. YNB (yeast nitrogen base) provides growth factors and inorganic salts for yeast growth, ammonium sulfate used as an inorganic nitrogen source, and sucrose is used as a fermentation carbon source. SS/-Ura with Agar is a mixture of YNB, DO Supplement-ra, sucrose, and agar powder, which uses the Ura defect in DO Supplement-Ura to screen strains that can express this component or to activate the relevant reporter.
