Rubisco-Bisphosphate Carboxylase/Oxygenase(Rubisco) Assay Kit
Rubisco-Bisphosphate Carboxylase/Oxygenase(Rubisco) Assay Kit
Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco, EC 4.1.1.39) is a key enzyme in plant photosynthesis, responsible for the fixation of CO2 while limiting the diversion of carbon to the Calvin cycle and photorespiration. The magnitude of its activity directly affects the rate of photosynthesis. Under the catalysis of Rubisco, one molecule of ribulose-1,5-bisphosphate (RuBP) combines with one molecule of CO2, producing two molecules of 3-phosphoglycerate (PGA); PGA can then be converted to glyceraldehyde-3-phosphate through the action of external 3-phosphoglycerate kinase and glyceraldehyde-3-phosphate dehydrogenase, accompanied by the oxidation of NADH to produce NAD. At 340 nm, NADH has a characteristic absorption peak, while NAD does not have this absorption peak; therefore, measuring the rate of decline in absorbance at 340 nm can represent the carboxylase activity of Rubisco.
