GSH is the most important antioxidant thiol substance inside cells, playing a significant role in antioxidant defense, protein thiol protection, and transmembrane transport of amino acids. The ratio of reduced to oxidized forms (GSH/GSSG) is a key dynamic indicator of the cellular redox state. Therefore, measuring the intracellular levels of GSH and GSSG, as well as the GSH/GSSG ratio, can effectively reflect the oxidative state of the cells. DTNB reacts with GSH to form a complex that has a characteristic absorption peak at 412 nm; its absorbance is proportional to the GSH content.
