pHHP-sgRNA-HsCas9-CENARS-dKU70 is a plasmid for Pichia pastoris CRISPR-9 gene editing. Its core feature is the use of the HXA gene promoter (HXA Promoter) to drive the expression of the Cas9 protein and the HB gene promoter (HXB Promoter) to drive the transcription of gRNA. dKU70 can shut down the NHEJ "noise" pathway, resulting in significant increase in the efficiency of homologous recombination editing.
