pJQ200SK vector
pJQ200SK was designed as a “suicide vector”, meaning that it cannot stably replicate in a broad range of gramnegative bacteria but can enter host cells. Its core functions are based on two key elements: “SacB gene negative selection”:The SacB gene is lethal to a variety gram-negative bacteria when expressed on a medium containing 5% sucrose. This allows cells that have successfully undergone gene replacement (i.e., lost the pJQ00SK vector) to survive on a sucrose plate, while cells that still retain the vector die, thus facilitating the screening of positive clones. “Conjugative transfer” The vector carries the “oriT”, which allows mobilization from one bacterium (e.g., a donor bacterium) to another recipient bium through conjugation.
