TPY agar medium is a solid medium for selective separation and enumeration of Bifidobacterium. Its basic components are casein peptone, plant peptone and yeast extract powder, providing a balanced nitrogen source and growth factors; The key additives are cysteine hydrochloride and mupirocin. The former is used as a reducing agent to create an anaerobic environment, while the latter inhibits most Gram-positive bacteria. Prebiotics such as fructooligosaccharides are specially added to the culture medium as the preferred carbon source, which can specifically promote the growth of Bifidobacterium and form typical colonies, and is widely used in the separation and quantitative analysis of Bifidobacterium in probiotic products and intestinal microbial research.
