In experiments such as plant genetic transformation, when a target plasmid (e.g., a plant expression vector) needs to be transferred from E. coli to Agrobacterium tumefaciens, pRK2013 can play a significant role if the target plasmid itself lacks mobility. It functions through aparental mating process: a helper bacterium carrying pRK2013, a donor bacterium with the target plasmid, and Agrobacteriumefaciens as the recipient are co-cultured. pRK2013 can temporarily provide the mating transfer function, acting as a bridge to help the plasmid efficiently enter Agrobacterium tumefaciens.
