Acid phosphatase decomposes disodium phenyl phosphate to produce free phenol and phosphoric acid. The phenol reacts with 4-aminoantipyrine in alkaline solution, oxidized by potassium ferricyanide to generate a red quinone derivative. The intensity of the red color can be used to determine the level of enzyme activity.
