DH5α
DH5α
DH5α is one of the most widely used classical cloned strains in Escherichia coli, and its main feature is its efficient DNA recombination defect. The mutation of recA1 and endA1 genes of this strain is its core advantage: the mutation of recA1 makes it lose its homologous recombination ability, thus greatly improving the stability of foreign plasmid DNA and avoiding unexpected rearrangement of inserted fragments; The mutation of endA1 inactivates its endonuclease I, which can produce plasmid DNA with higher quality and yield in small amount of plasmid preparation. In addition, DH5α also contains lacZΔM15 gene deletion, which can be used for blue and white spot screening with vectors such as pUC series to identify positive recombinant clones conveniently and quickly. Hsdr 17 (Rk–,MK+) in its genotype makes it a K-12-derived strain with limited system defects but complete modification system, which is beneficial to the transformation of foreign DNA. Because of these characteristics, DH5α has become the first choice host for plasmid construction, amplification, preservation and conventional gene cloning in molecular biology experiments, and it is favored for its high transformation efficiency, good stability and simple operation.
