EPI300 cells contain a mutant trfA gene, and the protein product expressed by this gene can promote the high-copy propagation of plasmids containing ori V replicon, and inducer I can induce the expression of trfA gene. Therefore, EPI300 strain can reduce the copy number of ori V replicon plasmid, which is especially suitable for cloning various unstable DNA or toxic genes. [mrr-hsdRMS-mcrBC] genotype makes EPI300 strain suitable for cloning DNA rich in methylcytosine or methyladenine. The mutation of recA1 and endA1 is beneficial to the stability of inserted DNA and the extraction of high purity plasmid DNA. The existence of lacZΔM15 marker makes EPI300 available for blue and white spot screening.
