SMD1163 is a protease-deficient engineering strain of Pichia pastoris, which is widely used in eukaryotic expression of recombinant protein. Its genotype is his4 pep4 prb1, which shows histidine auxotrophy and lacks the activities of proteolytic enzyme A(Pep4) and proteolytic enzyme B(Prb1). This double protease deficiency can significantly reduce the degradation of exogenous expressed protein by host protease, which is helpful to improve the yield and stability of target protein. The strain is of Mut+ phenotype, which can be induced by methanol normally. Usually, plasmid transformation is carried out by electrotransformation, and a vector containing histidine gene (such as pPIC9K) is used for compensation screening. SMD1163 is suitable for high-density fermentation culture, and the culture temperature is suggested to be controlled at 28-30℃.
