Y1HGold strain is a strain used in GAL4-AbA yeast hybrid system, MATα type, which can directly transform plasmid for screening library test. Transformation marker: ura3, leu2；; The reporter gene is AbAr. Y1HGold-GAL4-AbA yeast hybrid system needs two plasmids: pAbAi and PGADT7. The screening mark of plasmid pAbAi is URA, which is used to express pBait-AbAi construct (1~3 bait DNA sequences are cloned into pAbAi after repeated tandem); Plasmid pGADT7 was screened as LEU, which was used to express the fusion protein of AD (amino acids 768~881 at Gal4C terminal) and target protein (Prey). In addition, AbAr has the advantage of lower background compared with nutritional defect reporter gene, which can reduce the probability of false positive of yeast single hybrid.
