EGY48
EGY48
EGY48 strain is a strain used in LexA yeast two-hybrid experiment, MATα type, which can directly transform plasmid to verify protein interaction or screen library. Transformation marker are: his3, trp1,ura3, and the reporter genes are:LEU2;The reporter gene UAS (upstream activation sequence) is derived from LexA op(x6), and only when Bait and Prey LEU2 expression can only be started when interacting. EGY48-LexA system has three supporting plasmids: pLexA, pB42AD and p8op-LacZ. Plasmid pLexA is marked as HIS3, which is used to express the fusion protein of DNA-BD and target protein (Bait). The screening mark of plasmid pB42AD is TRP1, which is used to express the fusion protein of AD and target protein (Prey). The reporter plasmid p8op-LacZ was screened as URA3, the reporter gene was LacZ, and the reporter gene UAS was derived from LexA op(x6). Only when Bait and Prey interacted with each other could the expression of LacZ be started.
