Y1HGold Chemically Competent Cell
Y1HGold Chemically Competent Cell
The Y1HGold strain is a strain developed by Clontech for the GAL4-AbA yeast single hybrid system. It is of the MATα type and can directly transform plasmids for screening tests. The transformation markers are: ura3, leu2; and the reporter gene is: AbAr. The Y1HGold-GAL4-AbA yeast single hybrid system requires two plasmids to be used together: pAbAi and PGADT7. The selection marker for plasmid pAbAi is URA, used for expressing the pBait-AbAi construct (which consists of 1 to 3 bait DNA sequences cloned into pAbAi in tandem); the selection marker for plasmid pGADT7 is LEU, used for expressing the fusion protein of AD (the C-terminal 768-881 amino acids of GAL4) and the target protein (Prey). The principle of the GAL4-AbA yeast single hybrid system: Aureobasidin A (AbA) is a cyclic peptide antibiotic that can exert toxicity on yeast at low concentrations (0.1-0.2 µg/ml). The yeast strains integrated with pBait-AbAi (Bait-Reporter Yeast Strains) will activate the expression of AbAr when the prey protein binds to the bait sequence (Bait DNA), allowing it to grow on media containing the antibiotic AbA. AbAr has the advantage of lower background compared to nutrient-deficient reporter genes, which can reduce the probability of false positives in yeast single hybrids. The Y1HGold competent cells are processed by a special method and can be stored at -80°C for 12 months, with the pGADT7 plasmid (7988bp, AmpR) showing a transformation efficiency of >104 cfu/μg DNA.
